There are many ways to tweak the build for your usage, i.e get a smaller size SSD (120GB) and use the extra money for a dedicated GPU or downgrade to Haswell i3 (cheaper) then you can get a dedicated GPU while maintaining the SSD size, since you didn't mention HDD for storage, so I only include SSD.

Guys, I need some suggestions from you. I have a 2010 system which I think the motherboard just died on me.

Here's the existing spec,
I3-530 (LGA1156)
Gigabyte GA-P55-USB3L
2GB DDR1333
ATI 5750
3x HDD
Soundcard
ST50F (500W)
CM casing ATX Elite 334

As LGA1156 is not common anymore, I guess the next logical step is to replace the motherboard and CPU.

The question is, should I be looking at LGA1150 or LGA1151 now?

Quick check on the Viewnet price, here are my options.
A. If it's LGA1150, then it would be I3 4160 with H61 or H81 motherboard, or
B. LGA1151, I3 6100 with H110 or B150 motherboad.

The question here is, what is the cost effective way for me to enable the desktop up and running again? Appreciate your input.

I would say cost effective.

I think I have two options here,
A. The Haswell option, where this involves the CPU and motherboard,
B. The Skylake option, where this will involve the CPU, motherboard and RAM.

Is this right?
