Yes, you should drain the battery until iPhone shut down itself, then charged until 100% ( no need switch on, it will auto on once charged)

I'm using the latest iOS. Does that affect the battery consumption?

some claimed that the latest ios drained their battery. but for my iphone 5, the latest ios 6.1.2 doesnt have any battery drain problem. maybe you can restore the iOS, and set as new phone, reset network settings, drain the battery until the phone shut down, charge until full and then u can see the performance of the battery.
