Depending on the destination in Singapore.
Tuas to city center can take half an hour to one hour during peak periods. If the destination is western Singapore, get a bus that allows early alighting, and switch to a CausewayLink city bus at Tuas CIQ. Tuas to Boon Lay is ten minutes. No severe congestion expected. Congestion usually starts somewhere before Jurong Town Hall Road exit.
Tanjung Kupang to Tuas is affected by truck traffic, and occasionally bus traffic, at Tuas CIQ. Worst case in recent years is additional half an hour delay. Longest in history may be one hour.
Immigration clearance for tourists may add another half an hour.
TBS to Tanjung Kupang is usually affected by the congestion in KL and Selangor, also Negeri Sembilan, which can add another half an hour.
Without congestion, should be minimum 4.5 hours, unless the driver speeds. I would expect 5.5 hours as the norm.
