When the system was first proposed, Valve hinted that they were using a derived version of Microsoft's TrueSkill rating system (TSR). This system was originally developed for games such as Halo etc. As with most ELO-based systems, TSR is optimized for 1v1 games, and not the 5v5 game that DotA is (for such games, there needs to be a correlation between the impact one teammate has on another's rating). The reason TSR was selected as the base model to work with is it's high convergence rate, i.e. players get a relatively accurate rating within a small number of games. For 1v1 games, TSR can accurately rate a player in under 5 games. For a complex multiplayer game like DotA, this value inevitably increases (the actual minimum number of games to get a proper rating is unknown, but some people on Dotabuff have noted that they can get moved up to the next bracket by 6 games). As far as matching goes, we know that parties of similar size are preferable, and that each party is evaluated by it's average rating, with some bonus/penalty depending on the best player in the party as well as the party size. This indicates the use of a stripped down genetic algorithm to identify the optimum matchups. One inherent problem with genetic algorithms is to keep the matching accurate, it takes a considerable amount of time (i.e. server resources) to compute. In practice, a variable called the mutation rate is adjusted so that the matching becomes as accurate as possible for a given amount of time that's considered reasonable. This give and take is why Valve cannot please everyone - either matches are slightly unbalanced for certain members of the population, or it takes 30 minutes to queue. Overall, this presents a very complex mathematical problem, and it's one that's not going to be resolved quickly. Even the smallest of improvements take years to perfect.
