Have you tried doing a full restore? Maybe that could help out a bit. Otherwise, it could be hardware related. I remember last time when I got the first ever iPod Touch, it was defective. One part of the screen is dead (not unresponsive). It doesn't register my touches at all. Nothing can be done except for a swap.

If you're suspecting a calibration would resolve the issue, you should do a restore first. Cause its part of the process. Otherwise, you can try out those file uploading procedures.
