I have a Maxis plan (unlimited calls and SMS).
Just curious, if I reply to 5-digit SMS number (e.g. 66300) will there be any charges?
Does replying to 5 digit SMS cost money?

Depends on the destination short code if it is a subscription based service … usually short code only accept short answer for subscription purpose and most of the time cannot unsubscribe… so becareful.
