Try plugging in the usb stick and switch on the laptop. Certain laptops will boot straight to the usb. If it didn't ,try pressing the key shown during bootup to choose boot device. You can pull out the device once it finished installing the os. Safe bet will be when you type in your username password etc.
You don't have to initialise if you're gonna install a new os on a new drive. You can format the drive while installing the os itself. It will detect the drive like normal.
