Yes, surely it will.
In my old house, it took around 15 years ~ 20 years to have enough rust on the cover until holes start to appear. Back then I just made a cement based cover and cover it up. |

Cover with Sand mean it will block the drainage, and will give you nightmare.

Cover with bricks or decorative stones. Then put a potted plant on top of it. Like this you can still access the manhole when needed.
