First thing first, know the company inside out.
Understand the industry, competitors, products and services.
Then sell yourself:
1. Give them reasons why you should be hired (strengths etc.) 2. Your short and long term goals which should be relevant to the company. etc. 3. How you will be an asset that will augment the current, existing team.
Be confident and enthusiastic at the same time.
Show that you are passionate someone who is capable through your speech and body language.
Practice makes perfect.

it's something that's not so easy to practice... different HRs, different employers, there's a lot of psychology, thought what you really need is to reasonably access your skills, knowledge, and abilities to be absolutely confident while talking about them and also things you can do well on certain position

Not for a job per se, but during selection for specialist training, yes - I was interviewed by my direct supervisor, to see if he would endorse my application. When the application was endorsed, I was then interviewed by my head of department. Then, it got escalated to the Chairman of the Medical Board of my hospital, before moving upward to the Specialty Training Committee. The 3rd interview isn't one that normally occurs - but, because I wasn't a Singaporean, my HOD and the CMB wanted a commitment that I would not train and cabut after the training. So, in essence, that was 4 rounds of interviews... and gallons of sweat lost from the nervous sweating!

As long as you've kept your registration intact with the relevant authorities, you should have no issue restarting work. My education was more on Bioinformatics, earlier I was involved in healthcare line in Diagnostic Center. However, I left this line for quite some times but my passion has always been related to healthcare / medical. Was thinking of changing field but I am not really sure about current employment market in this line.

I personally feel that the healthcare sector (be it the front line staff like doctors and nurses, or those playing a supporting role - like diagnostic/lab personnel) is one of the more resilient sectors out there - resilient, not resistant to economic challenges. However, I really cannot comment on salaries, as I'm not in the exact same field as you are. I'm pretty sure that if you were to re-enter the medical field, you'd be more than welcome! Good luck!

Is there actually any portals or channels to apply for this line of work except for usual job portals? My impression on medical / healthcare line was people hiring mostly through referrals and connections.

Is it appropriate to use enquiry form on company website to follow up job application?
The HR contact is not listed on the job ad and I only have the general email which i applied the job jobs@XYZ.com, will I look silly if I use the enquiry form to follow up on such matter?
