Is d3 region locked? or is it something like WoW where you pick a realm or something like that?

latency affects pvp most.
A blue post mentioned that support for pvp will not be as strict and competitive as WoW and SC2 (source = http://us.battle.net/d3/en/forum/topic/4768348365?page=5#97) If your not into pvp than latency is fine for most. this coming from a short open beta play-through by yours truly so don't take my word for it. I am glad blizz is going in this direction. more time and development on the pve experience. PvP is indeed for fun only. No need so serious yo.
